In vitro asymbiotic germination of two endemic Chinese terrestrial Orchis seeds and the physiology of their germination response

摘要: Orchis diantha and O. chusua are endangered orchid species, and their seed germination is sparse in nature. To increase seed germination and protect these species, we successfully established the in vitro asymbiotic germination system of mature seeds.The results showed that 1/2 MS (Murashige and Skoog, 1962) medium supplemented with 6-benzyladenine or activated charcoal increased germination of O. diantha seeds, but had no promoting role for O. chusua seeds. In contrary, supplement of coconut milk to 1/2 MS medium increased the germination of O. chusua seeds, and had no role for O. diantha seeds. After-ripening marginally increased germination of O. diantha seeds, and significantly increased germination of O. chusua seeds. Light decreased germination of O. diantha and O. chusua seeds. There is clearly a difference in germination requirements between O. diantha and O. chusua seeds.
