Polymorphism of Single Sequence Repeat Markers Based on the Whole Genome Sequence of Agaricus bisporus and Its Use in Strain Identification

摘要: The white button mushroom Agaricus bisporus is the most widely cultivated species of edible mushrooms worldwide. Originating from a limited heritage line, commercial strains of A. bisporus are thought to be genetically very similar. Many highly polymorphic molecular markers have been exploited for strain identification in wild and commercial strains of this mushroom. Simple sequence repeat (SSR) molecular markers in the whole genome sequence, which are accurate, reproducible, and robust, are promising markers for genetic diversity analysis of many crops. In this study, 140 SSR molecular markers based on the whole genome sequence of A. bisporus strain H97 were found in 21 strains of mushroom collected from around the world. Polymorphic loci, represented by 37 distinct bands, were detected using 15 primer pairs. Clustering results showed that the 21 strains could be identified from 37 unique bands and divided into two groups of wild strains and cultivars. SSR primers successfully identified each individual. Our results demonstrate that SSR markers are powerful for detecting polymorphisms among closely related genotypes of A. bisporus.
