Csu-miR-375 and Csu-miR-11631 regulate ryanodine receptor to induce chlorantraniliprole resistance in Chilo suppressalis

摘要: Chilo suppressalis causes considerable rice production losses, and chlorantraniliprole is widely used for its management. Increased detoxification enzyme activities and point mutations in the ryanodine receptor (RyR), related to chlorantraniliprole resistance in C. suppressalis, have been reported. However, whether the up-regulation of CsuRyR is involved in chlorantraniliprole resistance and the corresponding regulatory mechanism mediated by microRNAs (miRNAs) are scarcely documented. In our study, 1.91-, 2.80-, 2.98- and 3.74-fold higher expression of CsuRyR got detected in four chlorantraniliprole-resistant populations (three field-resistant populations and a lab-resistant populations [SR], respectively) compared to a susceptible population (SS). Two miRNAs pairing with the 3'-UTR, Csu-miR-375, and CsumiR-11631, were predicted to target the CsuRyR gene, which showed an inverse expression pattern with CsuRyR in four chlorantraniliprole-resistant populations. Csu-miR-375 and Csu-miR-11631 were virtually transcribed in all the developmental stages and the tested tissues of C. suppressalis larvae but with higher expression in the SS population than in the SR population. Overexpression or knockdown of Csu-miR-375 and Csu-miR-11631 directly inverse regulated the CsuRyR expression and further impacted the mortality of C. suppressalis larvae caused by chlorantraniliprole. These results improve our understanding of the role of miRNAs Csu-miR-375 and Csu-miR-11631 in CsuRyR regulation and diamide insecticide resistance in C. suppressalis, providing new insights into resistance management strategies for controlling this destructive insect pest.
