Integrated metabolomic, transcriptomic, and phytohormone analyses revealed sucrose accumulation mechanism of muskmelon (Cucumis melo) during fruit ripening

摘要: Muskmelon (Cucumis melo L.) is a fast-growing, highly-valued horticultural crop prized for its taste. However, the regulatory mechanisms underlying sugar accumulation during the ripening process of muskmelon fruit remains unclear. In this study, during the late ripening of muskmelon fruit, sucrose accumulated to 65.26 mg g- 1, playing a more critical role than glucose (23.12 mg g- 1) and fructose (18.50 mg g- 1). The elevation in the sucrose level could be attributed to the heightened activities of sucrose phosphate synthase and sucrose phosphate phosphatase. Through metabolome analysis, a total of 877 differentially accumulated metabolites (DAMs) were identified across four different ripening stages, and pathway enrichment analysis revealed that the major DAMs were significantly enriched in 'Starch and Sucrose Metabolism'. Transcriptomic analysis identified 14,350 differentially expressed genes (DEGs), over 40 % of which were enriched in the 'Plant hormone signal transduction' and 'Starch and sucrose metabolism' pathways. In contrast to abscisic acid, auxin and ethylene were more crucial in sucrose accumulation. In addition, multi-omics integration and gene co-expression network analysis revealed a notable correlation between the accumulation patterns of crucial metabolites (glucose-1phosphate, uridine diphosphate glucose, sucrose-6-phosphate) and the expression trends of key functional genes, as well as those of potential transcription factors (CmERF022, CmNAC79, CmbHLH104, CmWRKY46). Our findings shed light on the molecular mechanisms and the crucial genetic factors that regulate sucrose accumulation in muskmelon fruit.
