Molecular tagging and mapping of a major linked marker for taproot thickness in a Brassica crop

摘要: A DNA marker linked to the locus controlling storage-root thickness was identified using bulked segregant analysis (BSA) in an F(2) population derived from a cross between turnip (Brassica rapa L. ssp. rapifera Matzg.) and rapeseed (Brassica rapa L.). Root-thickness in the F(2) plants showed a continuous distribution from 2.2 - 10.7 cm, suggesting the effects of a few major and minor genes. BSA, using three bulks based on root-thickness, suggested that an AFLP marker (E(ACC)M(CAG)-220) which appeared only in plants with a thickened root, was linked to a locus that controlled root size. This AFLP band was cloned, sequenced, and successfully converted into a simple, dominant, sequence characterised amplified region (SCAR) marker. This marker may help to identify turnip genotypes with thicker taproots in segregating progenies of root crops during marker-assisted selection (MAS) breeding programmes.
