Evaluation of elite inbred lines of maize (Zea mays L.) group Tangsipingtou for in vitro culture and plant regeneration

摘要: Tangsipingtou is the most important maize heterotic group in china since 1970. Knowledge of its potential in in vitro culture and plant regeneration is limited. in the present work, fifteen Tangsipingtou elite inbred lines were tried in vitro to evaluate the influence of genotype, medium, plant hormones and culture protocols on embryo-derived callus induction and plant regeneration. The results showed that callus cultures could be induced from all tested inbreds. Plant regeneration potential was reduced to a certain extent along with culture aging. The modalities of callus induction and plant regeneration of different genotypes were different allowing to the hormone type and concentration. Among fifteen inbreds tested, seven were best on medium CIdcb (modified N6 medium with 3.3mg/L dicamba), and inbred Jing24 was best on CI24d1(modified N6 medium with 1mg/L 2,4-D) for callus induction. However, RH7, J7, HXZ853, ZH81 and HyJC were found with high performance of both callus induction and plant regeneration and were suitable genotypes for genetic transformation.
