Cloning and Functional Analysis of TaHSP83 Involved in the Defence Response Against Puccinia Triticina in TcLr19

摘要: The objectives of this work were to obtain a TaHSP83 gene from the TcLr19 wheat line and understanding it's function in against the wheat leaf rust. A spliced sequence with the length of 1822 bp was obtained by rapid amplification cDNA ends (RACE), and the full length cDNA and genomic DNA of the TaHSP83 gene isolated from TcLr19, was 1619 bp and 4214 bp, respectively. TaHSP83 gene was located on cytoplasm, plasma membrane and nucleus by subcellular localization. Silencing of TaHSP83 in the leaf rust-resistant plants by virus induced gene silencing (VIGS) resulted in increased susceptibility to Puccinia triticina. Characterization of TaHSP83 may facilitate breeding for leaf rust resistance and better understanding of the evolution of the HSP83 genes in wheat.
