Determination of trichodermin in the metabolite of Trichoderma harzianum Rifai by gas chromatography-mass spectrometry

摘要: A method has been developed for the determination of Trichodermin in the fermentation metabolite of Trichoderma harzianum Rifai by gas chromatography tandem mass spectrometry. Ethyl acetate was the optimal solvent to extract the Trichodermin from the fermentation liquor. Trichodermin was identified with both retention time and the typical ion peaks at m/z 292, 277, 149, 135, 121 and 107, and its content in the sample was quantified with an external standard calibration curve method. Under the selected analytical conditions, the retention time of Trichodermin was 8.09 min, the limit of detection was 0.04 g/L and the linear relation was satisfied in the range of 0.5 to 50 g/L. The average recovery was 102.3% with RSD of 0.86%. The method was proved to be validated to determine Trichodermin in fermentation liquor samples of T. harzianum Rifai.
