Use of monoclonal antibodies for the serological differentiation of wheat and oat furoviruses

摘要: Until recently, the genus Furovirus included all the rod-shaped plant viruses transmitted by plasmodiophorid fungi but these have proved to be very diverse in genome organisation Recent proposals, which have been ratified by the International Committee on Taxonomy of Viruses, have revised the genus so that it now includes only those viruses that have two genome segments which are not polyadenylated and that do not have a triple gene block. The type member is soil-borne wheat mosaic virus (SBWMV), and oat golden stripe virus (OGSV) and sorghum chlorotic spot virus (SCSV) are also included. We have recently characterised isolates of wheat-infecting furoviruses from China and France and shown that these are both new members of the genus, which have been named Chinese wheat mosaic virus (CWMV) and European wheat mosaic virus (EWMV) respectively. The coat proteins (CP) of CWMV, SBWMV, OGSV and EWMV each consist of 176 amino acids with amino acid identities between them of 71% to 79%. The initial study showed that CWMV is serologically related to SBWMV. We report here the differentiation of these wheat and oat furoviruses by polyclonal antisera and monoclonal antibodies (MAbs) against (CWMV) and SBWMV.
