Production of haploid plants in wax gourd through parthenogenesis induced by gamma-irradiated pollen

摘要: Wax gourd is an economically important crop species in the Cucurbitaceae family, with most commercial varieties being F 1 hybrid. It is time-consuming to obtain homozygous lines by traditional breeding methods. Doubled haploid (DH) technology is an efficient approach to accelerating breeding process, which has been successfully used in various crop. Nevertheless, little is known about the study of haploid induction in wax gourd. In this study, successful haploid induction was achieved by induced parthenogenesis using pollen irradiated in wax gourd. The pollen of cultivar 'Rou2021 ' was irradiated with gamma ray at 100, 200, and 300 Gy. Female flowers were pollinated with irradiation pollen. The seeds extracted from the fruit were rescued on the culture medium for plant induction, and the ploidy level of the induced plants was determined by chromosome counting and flow cytometry. It showed that the number of fruit sets and induced plants was influenced by irradiation dose. Haploids were only obtained from lower irradiation doses at 100 Gy, with an efficiency of 0.13 to 0.14 %. In addition, there were significant differences in phenotypes and anatomical structures between haploids and diploids. Our results demonstrated for the first time that haploids were successfully induced using gamma ray irradiated pollen in wax gourd. The findings are of great value for the utilization of heterozygosity and the aggregation of superior traits in a short time, which significantly accelerate the breeding process and genetic analysis of wax gourd.
