Genome-wide identification of insulin-like growth factor-binding protein family in Chinese tongue sole (Cynoglossus semilaevis) and their responses to sex steroids

摘要: Chinese tongue sole (Cynoglossus semilaevis) is an economically important marine flatfish that shows typical female-biased Sexual Size Dimorphism (SSD) and sex reversal from female to pseudomale. Previous studies have shown that the insulin-like growth factor-binding protein (igfbp) genes are differentially expressed between sexes of C. semilaevis. The igfbp family is one of the important members in the IGF system, modulating somatic growth, development and reproduction. In the study, totally fifteen igfbp genes were obtained from C. semilaevis (designated as Csigfbps). Their open-reading frame sequences varied from 561 to 1131 bp and encoded 186-376 amino acids. According to phylogenetic and structural analyses, CsIGFBPs were clustered into ten groups and 6 of them were renamed. Interaction network prediction revealed that CsIGFBPs could interact with IGF1, IGF2, PAPPA, IGFALS, FN1a and other key molecules involved in growth and development. In addition, Csigfbp transcripts exhibited different abundances in pituitary, brain, gonad and liver from female and male C. semilaevis and most of these genes showed the highest abundance in male pituitary. The mRNA abundances of Csigfbp1a and Csigfbp3b were negatively associated with their hypomethylated ratios in the upstream areas, whereas the abundance and the hypomethylation within the gene body of Csigfbp8a showed a positive correlation. 17 beta-estradiol and testosterone differentially modulated the mRNA abundances of Csigfbps in brain cell lines. This study provided a foundation for further functional characterizations of Csigfbp genes and might be helpful to understand the growth and reproductive mechanisms of C. semilaevis.
