Study of an egg powder certified reference material for accurate measurement of florfenicol residue

摘要: A new egg powder certified reference material (CRM) was developed for florfenicol residue measurement. The incurred eggs were obtained from the laying hens that were orally administrated florfenicol. These eggs were freeze-dried, ground, sieved, homogenized, bottled and gamma-irradiated to prepare the CRM candidates. Liquid chromatography tandem isotope dilution mass spectrometry (LC-IDMS-MS) was established for the measurement of mass fraction of florfenicol in egg powder. The LC-IDMS-MS method was employed by all participating laboratories for characterization of florfenicol in egg powder CRM. The certified value was assigned to be 53.0 mu g/kg with expanded uncertainty of 6.9 mu g/kg (coverage factor k = 2, in the confidence level of 95%). The results of homogeneity testing show that the CRM was homogeneous for florfenicol between and within bottles. The long-term stability of the CRM at -20 degrees C storage condition was monitored for up to 13 month and short-term stability at 40 degrees C, 4 degrees C and room temperature for transportation and use was tested for nine days. The uncertainty was assessed by combing the contributions from characterization, homogeneity and stability. This CRM would be very useful for method validation and quality control of proficiency testing in the determination of florfenicol in egg.
