Molecular cloning and sequence characterization of common carp (Cyprinus carpio) integrin beta 1 (ITG beta 1) and its temporal expression in response to CyHV-3

摘要: Integrins, including the alpha and beta subunits, are cell adhesion factors that promote inter- cellular and extracellular matrix (ECM) interactions. Integrin beta 1 (ITG beta 1) is widely expressed in vivo. In addition, ITG beta 1 has been shown to promote virus entry into the body. Currently, common carp is severely affected by herpesvirus in the aquaculture industry. However, ITG beta 1 in common carp has not yet been identified. In this study, we cloned the full-length cDNA of CcITG beta 1 from common carp, which has an open reading frame (ORF) of 2394 bp and encodes a polypeptide of 797 amino acids, with a predicted molecular mass of 88.42 kDa and a theoretical PI of 6.018. Structural features of CcITG beta 1 were predicted by biological analysis software. Phylogenetic analysis showed that CdTG beta 1 of common carp was highly evolutionarily conserved and clustered with zebrafish ITG beta 1 alpha. CdTG beta 1 was found to be widely expressed in ten sampled tissues, with highest expression in the intestine. In fish selected to resist cyprinid herpesvirus 3 (CyHV-3), expression of CcITG beta 1 was significantly lower than that in control fish at different infection stages, especially the early infection stage. Thus, expression of CcITG beta 1 in common carp might play an important role in different stages of CyHV-3 infection, but further studies are required to verify this role.
