A novel Bacillus paramycoides HS-1 capable of degrading aflatoxin B1 and its key enzymatic mechanisms

摘要: Aflatoxin B1 (AFB1) is one of the major types of mycotoxins that endanger food safety, human health, and contribute to the immeasurable loss of food and agricultural production worldwide each year. In this study, a novel Bacillus paramycoides strain, HS-1, with a strong ability to detoxify AFB1, was isolated and characterized. HS-1 supernatant was confirmed to degrade 50.47 % of 2 mu g/mL AFB1 within 48 h at 30 degrees C. Furthermore, the supernatant exhibited a heat-activation characteristic; its degradation rate of 2 mu g/mL AFB1 increased to 77.17 % after heat treatment. The optimal conditions for the degradation of AFB1 by HS-1 supernatant were 70 degrees C and pH 11. Cu2+ slightly enhanced the degradation activity of HS-1 supernatant on AFB1. HepG2 cell proliferation assay further confirmed that AFB1 was transformed into substances with lower cytotoxicity after degradation by HS-1 culture supernatant. A novel AFB1-degrading enzyme FabI was identified from the HS-1 supernatant and could degrade 36.15 % of AFB1 within 48 h at 30 degrees C. The degradation products of AFB1 were identified as AFBO and AFB1-8,9-dihydrodiol. The characterization of AFB1 degradation by B. paramycoides HS-1 and the discovery of FabI provide a theoretical basis for analyzing the mechanism by which Bacillus spp. Degrade AFB1.
