RNAi-mediated silencing of nuclear hormone receptor 4 inhibits the development and reproduction in the 28-spotted potato ladybeetle, Henosepilachna vigintioctopunctata

摘要: The nuclear hormone receptor 4 (HR4), a key component of the NR6 subfamily, is integral to the 20-hydroxyec-dysone (20E) signaling pathway. This study employed RNA interference to explore the function of HvHR4 in the 28-spot-ted potato ladybeetle, Henosepilachna vigintioctopunctata (HVig), a major pest of solanaceous crops. Spatial-temporal expression profiling revealed that HvHR4 is predominantly expressed in the Malpighian tubules and peaks during the 1st instar larval stage. Silencing of HvHR4 in the 3rd instar larvae disrupted molting and caused abnormal new cuticle development, characterized by irregular bristle folding. RNAi treatment with 5 ng or 50 ng of dsHvHR4 induced 72% and 100% mortality rates, respectively. Gene expression profiling further demonstrated that HvHR4 knockdown suppressed critical 20E pathway genes, such as HvSAD, HvSHD, HvECR, HvUSP, and HvHR3, while upregulating HvSPO and HvE75. In the 4th instar larvae, HvHR4 silencing impaired new cuticle formation, leading to darkening of the old cuticle and reduced 20E titers. Additionally, RNAi-treated adult females exhibited significantly lower egg hatching rates, coupled with dysregulation of genes involved in egg development, including those associated with ion transport, immune response, and organic acid metabolism. These findings highlight the essential role ofHvHR4 in regulating development and reproduction in HVig, underscoring its potential as a target for RNAi-based pest control strategies.
