Dynamic changes of phytochemical profiles identified key points of flaxseed capsule maturation for lignan accumulation

摘要: Flaxseed is a rich source of lignans and hydroxycinnamic acids. In this study, the variation of lignans, hydroxycinnamic acids, and the transcription levels of genes involving lignan biosynthetic pathway of flaxseed capsules at different mature stages was investigated. Secoisolarisiresinol diglucoside showed a sharp increase during the early rapid growing period, with about a 24.7-fold enhancement till S3, followed a slower increasing rate and peaked at mature stage S6. The highest amounts of secoisolarisiresinol and phenolic compounds were observed at the initial stage S1. The secoisolarisiresinol diglucoside contents were clustered with the expression of key-coding genes involved in specific lignan biosynthetic pathway (including LuDPO, LuPLR, and (Lu)UGT), as well as the two genes related to phenylpropanoid pathway (including (Lu)CAD and (Lu)PAL). In contrast, the contents of hydroxycinnamic acids, coniferyl alcohol, and secoisolarisiresinol were clustered with LuCOMT, LuCCR, and (Lu)4CL, which were associated with phenylpropanoid pathway. Cluster analysis suggested that S3 was the key point during flaxseed capsule maturation for lignan accumulation. These results would provide theoretical guidance to promote cultivation of flaxseed capsules for lignan accumulation, which could be potential resource for pharmaceutical, cosmetic, and food applications.
