Brassinosteroid Affects the Elongation of Mesocotyl and Coleoptile in Zea mays L. by Regulating the Network of Circadian Rhythm under Severe Stress

摘要: The cooperative elongation both mesocotyl and coleoptile in maize (Zea mays L.) is associated with an ancient agriculture practice known as deep-seeding to improve its drought tolerance in germination. Circadian rhythm modulates diverse developmental processes throughout the maize life cycle, however, the relationships between circadian rhythm and mesocotyl/coleoptile elongation remains unclear. Herein, RNA-Sequencing (RNA-Seq) was constructed from mesocotyl and coleoptile in W64A and K12 at the seeding depth of 3 and 20 cm and 4.16 x 10(-3) M 24-epibrassinolide (BL) treatment. Totally 916 similar to 13499 differentially expressed genes (DEGs) in eight comparisons were respectively identified. GO and KEGG enrichment analyses identified the relevant biological functions and metabolic pathways of these DEGs. In addition, a diverse range of candidate genes were mainly obtained from plant hormone signal transduction, brassinosteroid biosynthesis, zeatin biosynthesis, phenylpropanoid biosynthesis, TCA cycle, starch and sucrose metabolism, glycolysis/gluconeogenesis, MAPK signaling pathway-plant, and peroxisome, Particularly, we also systematically analyzed the complex molecular mechanism network among 53 circadian rhythm-related DEGs. Finally, quantitative real-time PCR (qRT-PCR) of 17 selected DEGs was conducted to validate the RNA-Seq and gene differentially expression results. These findings provide valuable information for further elucidates molecular basis against deep-seeding stress and genetic improvement of deep-seeding tolerance in maize.
