Antibacterial mechanism of vitamin C against Staphylococcus aureus, reduction of toxicity of metabolites and its application in pasteurized milk

摘要: This study explored the antibacterial mechanism of Vitamin C (VC) against Staphylococcus aureus (S. aureus), reduction of toxicity of metabolites and its potential applications in pasteurized milk. VC inhibits the growth of S. aureus by damaging the cell wall, reducing intracellular ATP levels, inducing membrane depolarization, compromising membrane integrity, and altering cell morphology. The leakage of nucleic acids and proteins further indicates that VC disrupts the cell membrane. Electrophoretic analysis revealed that high concentrations of VC degrade genomic DNA and proteins. Crystal violet staining confirmed that VC inhibits biofilm formation. Real-time quantitative PCR analysis revealed that subinhibitory concentrations of VC modulated quorum sensing, virulence gene expression, adhesion, and global regulatory gene expression in S. aureus (P < 0.05). Hemolysis assays, Western blotting, and skin infection tests demonstrated that VC reduces the toxicity of S. aureus metabolites. Additionally, VC exhibited antibacterial effects in pasteurized milk, suggesting its potential as a natural preservative.
