Control of gummy stem blight in pumpkin using Streptomyces nigrescens NEAU-L66: Insights into its biocontrol mechanisms

摘要: Gummy stem blight (GSB), caused by Stagonosporopsis cucurbitacearum, is a prevalent disease threatening cucurbit production. To identify Streptomyces strains capable of controlling GSB on pumpkins, Streptomyces nigrescens NEAU-L66 was isolated from pumpkin rhizosphere soil and exhibited significant inhibitory activity against the mycelial growth of S. cucurbitacearum. The strain achieved a biocontrol efficacy of 78.05 % in pot experiments at a spore suspension concentration of 1.5 x 10(9) CFU mL(-1). Furthermore, S. nigrescens NEAU-L66 enhanced the activities of defense-related enzymes. Phenylamine lyase, peroxidase, catalase, and superoxide dismutase were increased by 90.97 %, 84.71 %, 116.47 %, and 96.04 %, respectively (p < 0.05), compared with disease treatment. The strain also promoted seedling growth by producing indole-3-acetic acid and degrading lignin, resulting in marked increases in plant height, root length, and biomass (fresh and dry weight). Additionally, the crude extract of S. nigrescens NEAU-L66 provided a 91.89 % control efficacy against GSB at a 5 % concentration. Biochemical analyses revealed that the extract disrupted the cell membrane integrity of S. cucurbitacearum, effectively inhibiting its growth. These findings suggest that S. nigrescens NEAU-L66 is a promising biocontrol agent for managing gummy stem blight in cucurbits.
