Isolation and Biological Characterization of Heart-Derived Mesenchymal Stem Cells from Goose

摘要: Objective: This research is designed to establish the system of isolation and culture for goose heart-derivedmesenchymal stem cells, explore its biological characteristics and differentiation ability in vitro. Methods: Heart-derived mesenchymal stem cells were isolated by enzymatic digestion. The specific surface markers of heart-derived mesenchymal stem cells were detected by reverse transcription-polymerase chain reaction, immunofluorescence and flow cytometry. Growth curve was drew to analyze cell viability. Oil Red O staining, alizarin red staining, alcian blue staining and reverse transcription-polymerase chain reaction assay were used to detect differentiation of the heart-derived mesenchymal stem cells. Results: Heart-derived mesenchymal stem cells were obtained successfully and sub-cultured to passage 22 in vitro. The expression of specific surface markers on cells were positive. The growth curve appeared in typical S curve. Furthermore, cells were induced to differentiate into adipocytes, osteoblast, and chondrocyte successfully. Conclusion: Heart-derived mesenchymal stem cells can be acquired from goose embryo heart and have shown strong self-renewal capacity and multipotential differentiation ability in vitro. The results denote that heart-derived mesenchymal stem cells can be an ideal seeded cells for clinical treatment and tissue engineering.
