PLANT REGENERATION FROM IN VITRO LEAF EXPLANTS OF PRUNUS PSEUDOCERASUS LINDL

摘要: Efficient shoot regeneration from in vitro leaf explants of fruit crops is affected by genotypes, basal medium composition, combination, and concentration of plant growth regulators. Regeneration ability of adventitious shoots from leaf explants of five genotypes of Prunus pseudocerasus Lindl. was compared. The effects of composition of basal medium, combination and concentration of plant growth regulators and sucrose concentration on shoot regeneration and rooting of adventitious shoots were examined. The effects of basal medium on adventitious shoot induction showed evident genotypic variability. For genotypes YC1 and YC3, all three tested basal media QL, WPM, and NN69 successfully induced shoot regeneration. For genotype YC4, basal media WPM and NN69 were more effective than QL for induction of adventitious shoots. For genotype YC2, shoot regeneration was obtained only on basal medium WPM. However, genotype YC5 failed to regenerate on all tested basal media and plant growth regulator combinations. The thidiazuron (TDZ) was more effective than 6-benzyladenine (BA) in inducing shoot regeneration for all studied genotypes. In vitro rooting capacity of adventitious shoots was genotype dependent and the highest rooting rate ranged from 31.1% to 98.9%. In general, adventitious shoot induction and rooting of the shoots of genotype YC5 was lower. The highest frequency of adventitious shoot induction of YC3 was obtained on QL medium with 2 mg l(-1) TDZ and 0.5 mg l(-1) IBA. The highest rooting rate of adventitious shoots was obtained on quarter strength MS medium with 0.5 mg l(-1) IBA.
