Isolation and Biological Characterization of Umbilical Cord Mesenchymal Stem Cells Derived from Goose Embryo

摘要: Mesenchymal stem cells observed in Wharton's Jelly of the umbilical cord (UC-MSCs) satisfy the criteria for MSCs and function as promising therapeutic candidates for clinical research. Nevertheless, their proliferation, differentiation and application potential have not been fully elucidated. In this study, we successfully established UC-MSCs line from 12 goose embryos samples and preserved 120 vials of cryogenically frozen cells. And, isolated UC-MSCs were expanded for passage 47 in vitro. The diploid ratio of the cells was over 92.7% (2n = 78) by karyotype analysis. Assessment of the growth curves showed typically sigmoidal. Moreover, UC-MSCs were observed to positively express cellular markers (CD29, CD44, CD71, CD90, CD105 and CD166) and the undifferentiated state marker (OCT4), but lack expression for hematopoietic cell markers (CD34 and CD45). In addition, multipotential differentiation of UC-MSCs was evaluated through differentiation into the specialized cells (adipocytes, osteoblasts, chondrocytes and neural cells) in vitro. Accordingly, UC-MSCs may play a significant role in cell regeneration therapy. Future study is further needed to investigate whether UC-MSCs are true stem cells through engraftment long-term in vivo.
