Multilineage Potential Research on Metanephric Mesenchymal Stem Cells of Bama Miniature pig

摘要: Objective: The objective of this research is to establish the system of isolation and culture for Bama miniature pigs's metanephric mesenchymal stem cells (B-MMSCs), and explore its biological characteristics and differentiation capacity. Methods: Bama miniature pigs's kidney was collected to isolate metanephric mesenchymal stem cells by enzymatic digestion, and then analyze its viability and detected the specific marker using sreverse transcription-polymerase chain reaction (RT-PCR), immunofluorescence and flow cytometry methods, Oil Red 0 staining, alizarin red staining, alcian blue staining and RT-PCR assay were used to detect its differentiation ability. Results: B-MMSCs could be cultured for 17 passages and had a strong capacity for self-renewal in vitro. Surface antigen detection showed that B-MMCs expressed CD90, CD133, Fibronectin, Nestin and Vimentin, but not expressed CD45. Furthermore, B-MMSCs could differentiated into adipocytes, osteoblasts and endothelial cells. Conclusion: The metanephric mesenchymal stem cell (MMSCs) could be isolated from Bama miniature pigs, and the cells possess powerful self-renewal ability and multiple differentiative potential. Therefore, the present study plays an important role by providing a metanephric mesenchymal stem cell choice for cell therapy and tissue engineering.
