Proliferation Potential and Biological Characterization o Muscle-Derived Stem Cells from Goose Embryo Skeletal Muscle In Vitro

摘要: Tissues repair after injury is a complex biological process, which involves the activation of tissue-resident precursors and stem cells and a variety of infiltrating cells responding to local and systemic signals. Skeletal muscle regeneration relies on the activation and proliferation of the resident muscle precursor cells including satellite cells and muscle stem cells. In this study, muscle-derived stem cells (MDSCs) were isolated from 26-day-goose embryo and primary MDSCs were subcultured to passage 37. The surface markers of MDSCs which including CD29, CD90, CD44, NCAM1 (CD56) and CD166 were detected by immunofluorescence and the RT-polymerase chain reaction assays. The proliferation potential of MDSCs were detected by the growth curves and colony forming assay. In addition, MDSCs at different passages were induced to differentiate into osteoblasts, adipocytes, chondrocytes and myotube successfully. The results suggest that the MDSCs isolated from goose skeletal muscle possess similar biological characteristics with those derived from other species, and their multilineage differentiation provides evidences for the development paradigm of the muscle stem cell, which would also provide theoretical basis for potential therapeutic applications of MDSCs in the treatment of muscle disease or injury.
