Identification and Biological Characterization of Mesenchymal Stem Cells from Bovine Synovial Membrane

摘要: Synovial mesenchymal stem cells (SMSCs) as a special type of Mesenchymal stem cells (MSCs), is likely to become a kind of seed cells to osteochondral repair. Previous studies mainly paid attention on humans and other model animals, but rarely performed on livestock. One aim of our research is to establish a systematic way which can successfully isolate and culture the bovine synovial mesenchymal stem cells (B-SMSCs) in vitro. The other aim is to explore the biological and differentiation functions of SMSCs. In our work, B-SMSCs were isolated from 8-week-old bovine embryo and studied on the cellular and molecular level. Primary SMSCs were subcultured to passage 30. The surface markers of SMSCs, including CD44, CD73, CD166, CD105 and CD90 were detected by RT-PCR assays and CD73, CD90, CD105, CD166 and CD44 were detected by immunofluorescence and flow cytometry. The colony forming efficiencies of different passages were tested. In addition, SMSCs were successfully induced to differentiate into adipocytes, osteoblasts, and chondrocytes. The results showed that the SMSCs which were successfully isolated from bovine embryo maintained the biological characteristics and the potential for multi-directional differentiation. Our research lays the foundation for further cell therapy of bovine osteochondral repair.
