Spatial metabolomics reveals the phytochemical basis of color variation and antioxidant capacity in quinoa (Chenopodium quinoa Willd.) varieties

摘要: Quinoa (Chenopodium quinoa Willd.) is a nutrient-dense pseudocereal renowned for its antioxidant properties, which vary significantly among white (WQ), red (RQ), and black (BQ) varieties. This study employed spatial metabolomics via MALDI-MSI to unravel the tissue-specific distribution of metabolites driving color-dependent antioxidant activity. Total antioxidant capacity exhibited a clear gradient (BQ > RQ > WQ) corresponding to seed pigmentation intensity, while spatial metabolomic profiling identified 2193 metabolites with distinct abundance among varieties. The comprehensive analysis revealed that traditional antioxidant compounds (flavonoids, phenolic acids, alkaloids, and saponins) were predominantly localized in embryonic tissues rather than the seed coat, contributing to bulk antioxidant activity but not fully explaining color-dependent differences. Instead, 81 seed coat-specific metabolites, including alkaloids (9), organic acids (27), and terpenoids (4), were identified as potential drivers of color-associated antioxidant variations. This unprecedented spatial resolution of quinoa's metabolome provides critical insights into the biochemical basis of its functional properties, enabling targeted breeding strategies and precision processing techniques to enhance bioactive compound retention in value-added quinoa products. The distinct spatial organization of metabolites further illuminates quinoa's evolutionary adaptations that simultaneously enhance nutritional value.
