In vitro and in vivo antiviral effect of molnupiravir against infectious hematopoietic necrosis virus

摘要: Infectious hematopoietic necrosis virus (IHNV) is the pathogen of infectious hematopoietic necrosis (IHN), which is a significant threat to the salmon and trout industry. To develop drugs that can inhibit IHNV, we screened a library containing 609 compounds. Molnupiravir, a nucleoside analogue, was found to effectively inhibit the in vitro infection of IHNV with EC50 of 5.554 mu M in this study. The targeting period of anti-IHNV effect was determined by the time of addition essay and the expression change of virus gene in attachment, internalization, genome synthesis, and release stages with drug treatment. The results showed that the molnupiravir targeted the early stages of the IHNV life cycle by affecting the attachment stage, and it also significantly decreased the specific infectivity (si) value of IHNV, a parameter evaluating the phenomenon of restricted virus/cell interaction. Molnupiravir was administrated through oral and intraperitoneal injections, respectively. The mortality rate caused by IHNV infection was significantly reduced in the fish that were intraperitoneally injected molnupiravir with a single dosage of 10 mg/kg (P<0.05). These findings suggest that molnupiravir has the possibility to be used as an antiviral drug for the prevention and control of IHN.
