An electrochemical biosensor based on a novel nucleic acid isothermal amplification method for detecting pathogenic bacteria

摘要: Detection of pathogenic bacteria is crucial to food safety. In this study, we established a novel nucleic acid isothermal amplification technology (Primer linear amplification, PLA) combined with electrochemical biosensors for ultra-sensitive and specific detection of pathogenic bacteria without the need for redundant nucleic acid extraction. In the presence of target E. coli O157: H7, the bacteria preferentially bound to the AP in the capture probe (CP) -aptamer (AP) double strand on the electrode, releasing the AP from the electrode. CP was re-exposed to the electrode, and the PLA-amplified repetitive long ssDNA bound to CP via base complementation. To achieve electrochemical signal transduction, methylene blue (MB) was incubated to generate electrical signals. The sensor could quantify E. coli O157:H7 concentration from 10 to 10(8) CFU/mL, with a detection limit of 6 CFU/mL. This method was effectively applied to sample analysis and could successfully detect pathogenic bacteria in tap water and milk samples.
