Cancer-targeted and biomarker-activated fluorescent probe for diagnosis imaging and therapy

摘要: Integrating the diagnostic and therapeutic processes can facilitate real-time detection and concurrent treatment. Despite the emergence of numerous high-performance fluorescent probes in recent years, most are specifically tailored for either the diagnosis or therapy, and the research on integrating dual functionalities for cancer remains limited. Herein, a fluorescent probe A-H responsive to adenosine triphosphate (ATP) and Hypochlorous acid (HClO) was successfully synthesized. It contained exceptional optical characteristics with high sensitivity and excellent selectivity in vitro assays, thereby enhancing the efficacy of cancer diagnostics and therapy. Because HClO and ATP are two vital biomarkers over-expressed in cancer cells, probe A-H could be used to distinguish normal cells from cancer cells for significantly bolstering the precision of cancer diagnostics. Upon entering cancer cells, A-H could interact with over-expressed HClO, releasing the critical agent methylene blue (MB) for photodynamic therapy (PDT). It was worth noting that the utilization of the fluorescent probe A-H has proven that combining ferroptosis and PDT not only overcame the limitations of single cancer treatment methods but also enhanced cancer therapeutic efficacy. Thus, probe A-H effectively integrated the functions of cancer diagnosis imaging and therapy, providing an effective strategy for developing integrated dual-functional fluorescent probes for targeted cancer diagnosis and treatment.
