Comparative transcriptome analysis identifies the MeNAC38-MeFRK4 module involved in sugar accumulation in cassava storage roots

摘要: Cassava (Manihot esculenta Crantz) is a globally important root crop, prized for its high starch content in storage roots. It is a valuable food source and a promising feedstock for bioethanol production. While sugar accumulation significantly impacts cassava yield and its industrial applications, the underlying molecular mechanisms remain largely unexplored. This study employed comparative transcriptome analysis and co-expression network analysis to identify key genes and regulatory elements involved in sugar metabolism in cassava storage roots. Using comparative transcriptome analysis, we identified MeFRK4 as a key gene influencing sugar accumulation, with its high expression in the low-sugar line (Y5) suggesting a negative regulatory role. Biochemical assays validated MeFRK4's fructokinase activity in vitro. Transcriptome-based co-expression analysis further revealed MeNAC38, a NAC transcription factor that directly regulates MeFRK4, confirmed through promoter analysis, structural modeling, dual-luciferases, and yeast one-hybrid assays. Our findings provide novel insights into the transcriptional regulation of sugar metabolism in cassava and highlight the MeNAC38-MeFRK4 module as a critical component of this process. This newly identified module presents a promising target for genetic improvement strategies to enhance cassava varieties for dual-purpose applications-optimizing high-sugar phenotypes for bioethanol production while developing low-sugar varieties for food security-thus advancing sustainable agriculture and bioenergy initiatives.
