The CYP392D8 gene is not directly associated with abamectin resistance, a case study in two highly resistant Tetranychus urticae strains

摘要: Overuse of the pesticide abamectin has resulted in many abamectin-resistant populations of the spider mite Tetranychus urticae, which is an important agricultural pest. Abamectin resistance has been predicted to be related to an overexpressed cytochrome P450 gene, CYP392D8. However, whether CYP392D8 truly contributes to abamectin resistance in T. urticae is unknown. To determine whether there is a functional link between CYP392D8-mediated metabolism and abamectin resistance, we cloned the CDS sequence of CYP392D8 and two variants among susceptible and resistant T. urtice populations were observed. We found that CYP392D8 was overexpressed in two abamectin-resistant populations and was induced following exposure to an LC30 concentration of abamectin. However, the heterologous expression and transgenic Drosophila melanogaster indicated that the two CYP392D8 variants could not metabolize abamectin and that enhanced expression of CYP392D8 was not sufficient to increase tolerance to abamectin. Taken together, our findings demonstrate that CYP392D8 does not contribute to abamectin resistance in two Chinese T. urticae strains directly.
