Micropropagation and Adventitious Shoot Regeneration of Healthy and Phytoplasma-Infected Jujube

摘要: Healthy and phytoplasma-infected shoot cultures of two jujube cultivars 'Jinsi 4' (Ziziphus jujuba Mill.) and sour jujube (Ziziphus spinosus Hu.) were selected as materials. In vitro shoots proliferation and adventitious shoot induction from leaf explants of healthy and phytoplasma-infected jujube were examined. Results showed that the proliferation media for healthy and phytoplasma-infected shoots of the same cultivar were obviously different. Proliferation of phytoplasma-infected shoots was betterthan that of healthy shoots. Phytoplasma-infected 'Jinsi 4' jujube and sour jujube both showed proliferation well on hormone-free MS medium, but healthy 'Jinsi 4' jujube showed no proliferation on hormone-free MS medium (MSO) and a little proliferation on MS medium with 1-4 mg/L benzyladenine (BA) and 0.4 mg/L indole-3-butyric acid (IBA). Healthy sour jujube couldn't proliferate on MSO, but proliferated very well on MS medium with 2 mg/L BA and 0.4 mg/L IBA. Shoot regeneration from leaf explants of phytoplasma-infected shoots was easier than that of healthy shoots. The highest shoot regeneration rates of two cultivars' diseased leaves were obtained on WPM medium with lower TDZ concentration of 0.1 mg/L. The highest regeneration rate of healthy sour jujube leaves was obtained on WPM medium with higher TDZ concentration of 1 mg/L, and healthy 'Jinsi 4' jujube leaves failed to regenerate. The possible reason of the difference of proliferation and regeneration between healthy and phytoplasma-infected shoots is discussed.
