Genome-wide identification of DREB1 transcription factors in Erianthus fulvus and the functional role of EfDREB1C in regulating cold tolerance of transgenic Arabidopsis and sugarcane

摘要: The high-altitude habitat of Erianthus fulvus (Saccharum fulvus) makes it a valuable germplasm resource for genetically enhancing the cold resistance of sugarcane. The C-repeat/DRE binding factor (DREB1/CBF) transcription factor (TF) family strongly affects the cold stress response of plants, but its characteristics and functions in Saccharum and related genera are still unclear. In this study, we detected 11 EfDREB1 genes from E. fulvus and analyzed their evolutionary relationships and structural characteristics. Seven EfDREB1 genes act as early cold-responsive genes whose levels are rapidly upregulated following 12 h of cold stress in E. fulvus. The cold-induced EfDREB1C gene encodes a nuclear localization protein that has a potent transcriptional self-activation function, and its promoter exhibits cold-induced characteristics. Homologous and heterologous expression of EfDREB1C promoted cold tolerance in both transgenic sugarcane and Arabidopsis. The results of the RNAseq analysis further indicated that the overexpression of EfDREB1C in sugarcane upregulated the expression of Ca2+ signaling genes, protein kinase genes, transcription factors, sugar metabolism genes, putrescine biosynthesis genes, cold-regulated genes (CORs), hormone signaling genes, and antioxidant-related genes. These findings offer important insights into the cold stress adaptation mechanism of E. fulvus and establish a foundation for future applications to genetically improve the cold resistance of sugarcane.
