Characteristic analysis of heat-shock-cognate 70-4 gene promoter from silkworm, Bombyx mori (Bmhsc70-4)

摘要: Heat shock cognate proteins (HSCs) are homologous to heat shock proteins (HSPs) and constitutively expressed. HSCs facilitate correct protein folding and translocation and could be influenced by a variety of extracellular signals. The 1.4 kb long hsc70-4 promoter from Bombyx mori (Bmhsc70-4) was amplified and cloned into the plasmid for driving the luciferase reporter gene. Transient expression assays were carried out to investigate the effects of heat shock treatment, developmental stages, insect ecdysone and juvenile hormone analogue (JHA) on the Bmhsc70-4 promoter activity. The results demonstrate that the Bmhsc70-4 promoter activity was high in BmN cells, Sf21 cells and silkworm larvae at normal temperature. By heat-shock treatment at 37 degrees for 2 h, the Bmhsc70-4 promoter activity increased by 4.40-, 16.12-, 3.11-folds to the controls in BmN cells, Sf21 cells (Spodoptera frugiperda) and 4th instar silkworm larvae, respectively. But heat-shock treatment for 5th instar silkworm larvae led to a drastically decrease of the activity. Transcriptional activity of promoter was changed with the different developmental stages and reached the highest level at the wandering stage in 5th instar. Ecdysone treatment had no significant effect, while JHA treatments showed typical dose-dependent effects on the Bmhsc70-4 promoter activity in BmN cells. Low level of JHA (1 mu g/ml) increased the Bmhsc70-4 promoter activity, but high level (>5 mu g/ml) decreased its activity. In 5th instar silkworm larvae, spreading of high level JHA (>150 mg/kg) or injection of low level of ecdysone (1 to 3 mu g per os) increased the Bmhsc70-4 promoter activity. From the above results obtained from in vitro and in vivo, we can deduce that the Bmhsc70-4 promoter is both constitutive and inducible promoter. Due to its high transcriptional activity and inducible by the environmental signals, this implies that Bmhsc70-4 promoter is an ideal candidate in application of transgene or cell stable expression system in Bombyx mori.
