Antiviral activity of arctigenin against Chinese rice-field eel rhabdovirus in Monopterus albus

摘要: Chinese rice-field eel rhabdovirus (CrERV) causes serious disease and leads to heavy economic losses in Chinese rice-field eel farming industry. There is currently no established treatment strategy for CrERV. We evaluated the antiviral properties of 8 natural compounds against CrERV using Gibel carp brain (GiCB) cells. Among these compounds, arctigenin showed substantial inhibition of CrERV replication, reaching the highest inhibition rate of 63.90 +/- 3.93 % at 2 mg/L in GiCB cells. Arctigenin preserved the normal morphology of CrERV-infected cells, reduced the intracellular nuclear damage, and disrupted CrERV infection by interfering with viral attachment and entry. Time of addition studies suggested that arctigenin might exert its antiviral effect during the initial phase of virus replication. In vivo experiments demonstrated a significant 33.33 % increase in the survival rate of Chinese rice-field eel upon arctigenin administration. In summary, arctigenin has been verified as a natural substance possessing exceptional anti-CrERV activity, showing promise for potential use in aquaculture treatment of CrERV.
