THE BMSPATZLE5 GENE ENCODES A COMPONENT OF TOLL-SIGNALING PATHWAY AGAINST FUNGUS AND GRAM-POSITIVE BACTERIA INVASION IN THE INTEGUMENT OF SILKWORM, BOMBYX MORI

摘要: Spatzle (spz) plays an important role in the innate immunity of insects. In this study, the Open Reading Frame(ORF) of Bmspz5 gene and its 3 'end non-coding region were cloned from Bombyx mori (B. mori). The gene encodes a protein of 405 amino acids, of which 56 amino acids with the C-terminal fragment were shown to be different from the corresponding predicted ORF in silkworm genome database. Phylogenetic tree and sequence analyses showed that Bmspz5 has a common conserved Cysteine Knot region and is closely associated with spz5 of several other insects. The expression of Bmspz5 was upregulated in the integument 24 h after Yeast and Bacillus subtilis invasion, In addition, RNAi knockdown of Bmspz5 caused an obvious reduction in the transcription expression of the transcription factors Cactus, Rel and in the antimicrobial peptides Attacin, Moricin, and Gloverin in the integument of B. mori. These results indicate that Bmspz5 participated in the Toll signal transduction pathway in the integument of B. Mori.
