Effect of Auxins on In Vitro Propagation of Sour Jujube Infected by Witches' Broom Phytoplasma

摘要: The effects of varying levels and different auxins on in vitro culture of sour jujube shoots infected with witches' broom phytoplasma were studied. Number of infected shoots multiplication was increased with the increase of 3-indoleacetic acid (IAA) concentration. The maximum number of infected shoots proliferation reached 17.4 when cultured on medium containing 4 mg/L IAA, and maximum multiplication number of healthy shoots was 4.6 on medium containing 2 mg/L BA and 0.4 mg/L IBA. And 3-indolebutyric acid (IBA) and alpha-naphthaleneacetic acid (NAA) at different concentrations from 0.5 to 4 mg/L did not affect on proliferation of phytoplasma infected shoots, strongly indicating that witches' broom phytoplasma caused changes in endogenous cytokinin and auxin levels, and may cause cytokinin increase, because the better propagation of in vitro shoots required the proper ratio of cytokinin and auxin. Supplement of exogenous IAA promoted in vitro shoot proliferation of phytoplasma infected jujube, supported previously reported that the increase of zeatin concentration of diseased jujube resulted in jujube witches' broom symptom.
