Photoperiod-regulated circadian clock genes regulate reproductive diapause in Coccinella septempunctata

摘要: Many organisms have evolved circadian clocks to adapt to daily environmental changes, and utilize photoperiodism to regulate seasonal events such as diapause. Although it is generally accepted that similar molecular mechanisms govern both circadian and seasonal rhythms, conclusive evidence for such a system in insects remains lacking. In this study, we investigated this issue using RNA interference (RNAi) in Coccinella septempunctata. First, we identified key clock genes within the C. septempunctata genome and examined their corresponding protein structures and molecular evolutionary relationships. Second, RT-qPCR analysis revealed significant changes in the expression of several clock genes, including period (Csper), timeless (Cstim), clock (Csclk), cycle (Cscyc), and PAR-domain protein 1 (Cspdp1), between diapause and non-diapause conditions. Third, when C. septempunctata were transferred from long-day to short-day conditions, RNAi of specific clock genes, such as Csper and Cstim, resulted in alterations in ovarian morphology and lipid accumulation. Our findings suggest that these circadian genes do not directly affect ovarian development, but likely participate in upstream photoperiodic pathways that regulate diapause initiation. These results provide new insights into the complex role of circadian clocks in the regulation of insect diapause and reproduction.
