Isolation and biologic characteristics of a serotype 1 Marek's disease virus from chickens in Fuzhou

摘要: A field isolate of Marek's disease virus (MDV), designated Fl, was identified as serotype 1 MDV by Immune-fluorescence assay with type specific monoclonal antibodies. On primary isolation, Fl virus grew slowly in Muscovy duck embryo fibroblast (MDEF) cell cultures. In early stages of infection in MDEF, cells became round and more refractive than neighboring cells. Later, the cytoplasm membrane of these infected cells was changed so as to fuse themselves into one large syncytium, Paralysis and neuritis were observed in 53% of susceptible day-old chickens inoculated with MDV Fl at a low cell culture passage level in MDEF. Pathogenicity of the Fl strain was lost as a result of long cell-culture passage in chicken embryo fibroblast (CEF) cell culture. Chickens inoculated at Day-old with the F1 strain were resistant to the oncogenic effect of subsequent challenge with virulent MDV Beijing strain at 1-week-old. These promising features suggested that F1 strain should be a excellent candidate of attenuated vaccine virus to Marek's disease.
