Establishment of Adventitious Bud Regeneration System from Bulb of Muscari Botryoides Mill

摘要: The bulblets of Muscari botryoides Mill were selected as explants to develop the system of incubation in vitro on MS medium. Different sterilizing duration, various hormone as well as sucrose concentrations were designed for observing changes of the explants. The results show that the suitable sterilizing method of explants is to sterilize the scales for 8 minutes with 0.1 % HgCl2 solution generating that the contamination rate is only 4 %. The optimum combination for callus induction is MS medium with the addition of 2 mg/L 6BA, 0.5 mg/L NAA and 5 % sucrose. Dark condition is conducive to callus induction. The optimal proliferation medium for adventitious buds is MS supplemented with 2.0 mg/L 6BA, 0.1 mg/L NAA and 3 % sucrose. The appropriate rooting medium is MS in combination with 0.3 mg/LNAA and 3.0 % sucrose. The survival rate of transplant is 95 % when the seedlings are transplanted into the matrix containing peat, vermiculite and soil with volume ratio 2:1:1:2.
