摘要: 载脂蛋白B mRNA编辑酶催化多肽3F(APOBEC3F)是有效阻止内源性与外源性反转录病毒复制的新型宿主细胞因子。本文对猪APOBEC3F基因外显子6序列进行扩增和测序,筛选多态性位点,并分析其与PRRSV易感性的相关性。测序发现外显子6存在3个单碱基突变(g.337 T/G、g.341 G/A和g.343 T/C)。针对g.341 G/A建立CRS-PCR-RFLP方法,检测9个猪群193个样本,发现仅在姜曲海猪和梅山猪中有GG(0.79/0.80)和GA(0.21/0.20)基因型,其他7个猪群体只检测到GG基因型。GG基因型猪肺泡巨噬细胞中PRRSV拷贝数在接毒后18h显著高于GA基因型个体(P<0.05)。表明APOBEC3F基因外显子6与猪种PRRSV抗性相关,外显子6的341位点A等位基因更有利于抵抗PRRSV的感染,这为进一步探讨APOBEC3F基因作为猪种PRRSV抗性辅助选育标记提供了试验依据。
