猪源Viperin蛋白抑制猪繁殖与呼吸综合征病毒感染MARC-145细胞

摘要: 为了研究干扰素刺激基因Viperin对猪繁殖与呼吸综合征病毒的抗病毒作用,本研究构建了猪源Viperin基因(sViperin)pCI-sVIP表达载体,并在MARC-145细胞上观察了其对PRRSV复制的抑制作用。结果为:过表达sViperin蛋白可以抑制PRRSV在MARC-145细胞上的复制,且具有剂量依赖作用。sViperin蛋白抑制了病毒的入胞,但其对病毒的装配和释放没有影响作用。共聚焦实验证明sViperin蛋白主要定位于内质网等细胞器。sViperin蛋白和PRRSV GP5、N蛋白在细胞内存在共定位现象。CO-IP实验证明sViperin蛋白可以和PRRSV N蛋白存在相互作用。该研究为该病毒新型抗病毒药物研究奠定了重要基础。
