Combined transcriptomic and metabolomic analysis reveals the regulatory mechanism of pumpkin rootstocks on fruit quality of grafted cucumbers

摘要: Grafting is an advantage strategy for cucumbers to cope with various adversities, but the influence of grafting on fruit quality and its underlying mechanism remain to be fully explored. Here, non-grafted cucumbers served as the control (YN-S), while black-seeded pumpkin (B-45-G) and yellow-seeded pumpkin (Y-65-G) were utilized as rootstocks to investigate the influence of grafting on cucumber fruit quality and underlying mechanism. Results showed that Y-65-G fruits had superior taste and nutritional qualities compared with B-45-G and YN-S fruits. Specifically, Y-65-G fruits were sweeter, with higher soluble solid and soluble sugar contents, while B-45-G fruits exhibited a slightly bitter taste and reduced Vc content and cucumber flavor. Both grafting treatments increased the content of health-promoting secondary metabolites in the fruits, thereby enhancing edible value of the fruits. Nevertheless, more flavonoid compounds were accumulated in the Y-65-G fruits, while more lipid compounds, particularly Lysophosphatidylethanolamine and Lysophosphatidylcholine, were accumulated in the B-45-G fruits. The elevated sweetness in Y-65-G fruits was correlated with the increase in six sugar compounds. In contrast, the increased bitterness in B-45-G fruits was associated with multiple factors, especially the increase in polyphenols and alkaloid compounds. The genes coding for cinnamyl-alcohol dehydrogenase and shikimate Ohydroxycinnamoyltransferase highly contributed to the accumulation of flavonoid compounds in Y-65-G fruits, while phospholipase A, choline-phosphate cytidylyltransferase and lysophospholipid acyltransferase were obiviously related to the accumulation of Lysophosphatidylethanolamine and Lysophosphatidylcholine in B-45-G fruits. Therefore, to improve cucumber fruit quality, appropriate grafting rootstocks need to be carefully selected.
