Physiological and transcriptomic analysis of salicylic acid on membrane lipid metabolism and cell wall metabolism in postharvest peach fruit

摘要: Peach fruit is susceptible to postharvest softening during low temperatures, which significantly reduces fruit quality and commercial value. The mechanism of peach fruit softening mediated by salicylic acid (SA) is unknown. This study aimed to investigate the effect of SA treatment on membrane lipid metabolism and cell wall metabolism of peach fruit during cold storage. The results showed that SA maintained lower relative electrical conductivity, lower total soluble solids and reducing sugar content, and higher titratable acid content. SA treatment delayed the decrease of phosphatidylcholine, phosphatidylethanolamine, linoleic acid, linolenic acid and unsaturated fatty acids /saturated fatty acids (USFA/SFA) ratio, while reducing the increase of palmitic acid, stearic acid, and oleic acid. Additionally, SA treatment inhibited lipoxygenase, lipase and phospholipase D activities, down-regulated PLA genes and some LOX genes and up-regulated PAP genes in lipid metabolism pathways. Furthermore, SA treatment reduced pectin methylesterase (PME), polygalacturonase (PG), R-glucosidase (R-Glu), R-galactosidase (R-Gal) and Cx-cellulase activities and soluble pectin content, slowed the decline in cellulose and protopectin content, and reduced the expression of PME, PG, and pectin lyase (PL) genes. Therefore, SA treatment could prolong storage time by regulating membrane lipid metabolism and cell wall metabolism.
