Drought-growth promoting bacteria enhance drought tolerance by improving root conformation of Caragana microphylla

摘要: Plant growth-promoting rhizobacteria (PGPR) are known for their capacity to bolster plants' resilience against arid conditions. However, the underlying mechanisms remain unclear as to how the drought-tolerant PGPR strains modulate the drought resistance of inherently drought-resistant plants under water-deficit stress. This study simulated drought conditions using PEG6000 (polyethylene glycol) and employed bacterial separation and cultivation techniques to construct a drought-resistant and plant growth-promoting bacterial community. A twofactor (drought stress x drought-resistant bacteria) randomized block trials were used to investigate the effects of this community on the growth of Caragana microphylla plants, soil properties, and microbial communities. The major findings show that under drought stress, inoculation with PGPR significantly increased the plant growth indicators such as root length, surface area, volume, connections, nodes, root tips, branching numbers, and root biomass. The results show PGPR also significantly increased the nutrient availability such as the increase in the phosphorus content in the soil, nitrate nitrogen content, and alkaline phosphatase activity while reducing the pH in the soil. The findings further indicate that under drought stress, inoculation with PGPR significantly increased the diversity of soil microbial communities, altered the structure of soil microbial communities, and significantly enriched populations at the genus level such as Bacillus, Pseudarthrobacter, Iamia, and Massilia. This study confirms that under drought stress, the drought-resistant PGPR improves the microenvironment of Caragana microphylla's rhizosphere and regulates the root architecture, thereby promoting the growth of its roots and enhancing its drought resistance. These findings clarified the mechanisms of how the soil microbial communities planted with Caragana microphylla respond to drought and enhance the stability and functionality of artificial vegetation of Caragana microphylla.
