摘要: The commercialization of Antarctic krill oil raises concerns about product authenticity and adulteration. In this study, we developed a DNA-based authentication method for Antarctic krill oil. Species-specific PCR primers targeting a 157-bp fragment of the Euphausia superba 16S rRNA gene were designed, enabling high-specificity differentiation from Thunnus alalunga, Gadus ogac, and Salmo salar. The optimized PCR method achieved a detection sensitivity of 0.1 ng DNA/reaction. To address challenges in DNA extraction from complex lipid matrices, n-hexane and choline-based deep eutectic solvents (DES) were screened for their ability to separate interferents and enrich DNA. Among these, choline chloride-formic acid was selected as the optimal solvent due to its broader positive electrostatic potential (ESP) distribution (108.7 & Aring;(2), 56.0%). Post-PCR analysis successfully distinguished Antarctic krill oil from other edible oils. This work provides a robust tool for protecting high-value oils and advancing authenticity verification in related products.
