ISOLATION AND ANALYSIS OF RESISTANCE RELATED GENES IN WHEAT INDUCED BY FUSARIUM GRAMINEARUM BY SUPPRESSION SUBTRACTIVE HYBRIDIZATION

摘要: Fusarium head blight (FHB), caused by Fusarium graminearum, is a widespread and destructive disease of wheat worldwide. Currently, the molecular mechanisms of resistance in wheat to FHB had been focused on QTL or molecular mapping. In order to investigate the genes related to FHB resistance, a forward subtractive cDNA library was constructed using Sumai 3 and its susceptible near-isogenic lines as tester and driver, respectively. A total of 100 positive clones were picked randomly and sequenced. Based on the clustering result, the ESTs could be grouped into 64 contigs. The size of the ESTs ranged from 106-643bp, and the mean value was 274bp. Fifty-three of the ESTs showed similarity to known protein sequences in GenBank, the other eleven ESTs demonstrated to be novel sequences. The functions of the 53 ESTs were related to metabolism, energy, disease/defence, transcription and so on. The most prevalent categories were energy and primary metabolism, which accounted for 26.4% and 20.7% of the total ESTs, respectively. About 13.2% of the ESTs had relationship to disease/defence related genes, such as glyoxalase I, catalase isozyme, cytochrome P450, glutathione S-transferase, chalcone synthase and so on.
