Transcription factors StABR1 and StbHLH074 participate in starch and sugar metabolism in potato (Solanum tuberosum L.) during the storage and rewarming process

摘要: Potatoes (Solanum tuberosum L.) stored under low-temperature conditions tend to exhibit an undesirable sweetening. This is attributed to the degradation of starch into reducing sugars. Conversely, the rewarming process rapidly reduces the sugar content. Although (3-amylase (BAM) and acid-invertase (INV) have been extensively investigated during low-temperature storage, their transcriptional regulation remains rarely studied. In our study, the expression of sweetening-related genes StBAM1 and StINV1 in potatoes was induced during cold storage and inhibited during the rewarming process. Additionally, two transcription factors, StABR1 and StbHLH074 (selected from previous RNA-Seq data), exhibited expression patterns similar to those of StBAM1 and StINV1. The GUS assay demonstrated that StABR1 and StbHLH074 enhanced the expression of StBAM1 by increasing its promoter activity. The yeast one-hybrid assay revealed that StABR1 and StbHLH074 bind to the StBAM1 promoter and interact with each other. Furthermore, transient over-expression of StABR1 and StbHLH074, respectively, upregulated the expression of NtBAM1 and NtINV1, leading to decreased starch content and increased sugar content in tobacco leaves. It was concluded that StABR1 and StbHLH074 participate in starch and sugar metabolism in potato during the storage and rewarming process by regulating starch and sugar- related gene expressions.
