Effect of culturing factors on the production of biomass and secondary metabolites in ginseng adventitious root cultures for new resource food

摘要: Ginseng (Panax ginseng C. A. Meyer) adventitious roots (ARs) were cultured with various nutritional factors to determine optimal conditions of macro and microelements for root biomass in solid medium and a flask. Growth characteristics, including the number of lateral roots per explant, length of lateral roots, fresh weight and dry weight, were tested to understand the effect of nutritional factors. The optimized conditions for the AR growth were 0.75 x Murashige and Skoog (MS) with an ammonium and nitrate ratio of 7.5:22.5 mM and 1.0 x MS of microelements in solid and flask cultures. On the basis of these investigations, bioreactor cultures were established, and chemical constituents and the antioxidant potential of extract of ginseng ARs were studied. Accumulation of 1.13% total saponin and 20.9% total polysaccharides were achieved with ARs grown in 5 l bioreactors. Inorganic elements analysis revealed that ginseng AR contains abundant major elements and trace elements. Moreover, copper, lead, mercury and arsenic were within the limit range of health standards in food. The nutritional component data indicate that ginseng AR is rich in nutrition and low in energy. Unlike the concentration for 50% of maximal effect (EC50) of 1,1-diphenyl-2-picrylhydrazyl scavenging in ARs and field-grown plants, the EC50 of the extracts in ARs was much lower than that in field-grown plants. For the two groups of ginsenosides (Rb and Rg), the amounts of Rb and Rg2 that accumulated in the ARs were higher than those accumulated in untreated cultures after elicitation with 100 mu M methyl jasmonate (MJ) in a two-stage bioreactor culture method. Therefore, the AR of ginseng tissue culture has the characteristics of high-quality ginseng food, and the MJ elicitation in the two-stage culture was highly effective for enhancing ginsenoside accumulation.
