Neglected contributions of residual herbicides at various levels on soybean root rot: link with herbicide non-targeted soil microbial communities and functions

摘要: Multiple residual herbicides persist in soil of soybean-producing areas, posing a continuous risk to soil ecosystem. While the understanding of combined herbicide effects on soil/plant health related specific microbial communities (e.g., soybean root rot pathogens) remains limited. Herein, we conducted a pot experiment and in-vitro validation experiments, identifying microbial and functional variations under stress of combined herbicides at various residual levels. Combined herbicides at high residual level negatively affected soybean growth and significantly contributed to soybean root rot (P < 0.05). Microbial complexity and stability were significantly reduced by herbicides at moderate and high levels, conducive to pathogen invasion. The typical pathogens of Fusarium spp. exhibited as the keystone taxa in microbial networks or significantly (P < 0.05) enriched in treatments with herbicide residues. Potential pathogen-resistant bacteria and key microbial functions decreased under stress of herbicides (e.g., Candidatus Solibacter, RB41 and Vicinamibacteraceae). Key microbial functions (KEGG pathway annotations) of nervous system, digestive system, biosynthesis of secondary metabolites and glycan biosynthesis were also negatively affected by herbicides based on functional profile, indicating reductions in overall disease-suppressive functions of microbial communities. Additionally, herbicide residues affected soil properties and potentially created unfavorable conditions for survival of pathogen-resistant bacteria, thereby increasing the occurrence of soybean root rot. These findings provide improved evidence regarding the effect of combined residual herbicides on soybean root rot, with direct facilitation at high level and potential contributions at low and moderate levels, proposing new insights into soil environmental management and microbial regulations for strengthening disease-resistant ability of microbial communities.
